Evaluación de dos diluyentes seminales para la preservación refrigerada de espermatozoides de gato (Felis catus). Nota técnica

Con el objetivo de evaluar el efecto de dos diluyentes seminales en la preservación a 4° C de espermatozoides de gato, 10 eyaculados fueron obtenidos mediante vagina artificial. Aleatoriamente se asignaron 5 eyaculados por cada diluyente en estudio. El semen se evaluó inmediatamente y se diluyó en relación 1:5 con diluyente EYT (a base de yema huevo y buffer Tris: 510 mOsm/L y pH 7,5) o diluyente TEST (a base de yema de huevo y buffer Tris y Tes: 555 mOsm/L y pH 7,7). A continuación se evaluó la motilidad progresiva (MP) y vitalidad espermática (VE). Cada eyaculado una vez diluido, fue mantenido por 30 minutos a 25° C y luego enfriado a una tasa de 0,33° C/minuto hasta alcanzar 5° C y entonces almacenado en refrigerador a 4° C. Las evaluaciones de MP y VE se realizaron a las 24, 48, 72, 96, 120 y 144 horas de refrigeración. La MP fue superior (P<0,05) en el diluyente EYT a partir de las 24 horas de evaluación, alcanzando valores sobre 50 % a las 72 horas de refrigeración. La VE no presentó diferencias significativas al comparar los diluyentes en los diferentes tiempos, sin embargo se observó un descenso lineal a través del tiempo. Considerando que ambos diluyentes contenían yema de huevo y que la principal diferencia en composición entre ambos radicó en el tipo de buffer; las diferencias observadas en la motilidad progresiva, podrían ser atribuidas principalmente a las propiedades físicas de los diluyentes en estudio. En conclusión, es posible preservar espermatozoides de gato, refrigerados a 4° C, por períodos de hasta 72 horas, con un diluyente a base de yema de huevo y Tris.

Five ejaculates were randomly assigned to each extender under study. The seminal volume was evaluated immediately and the semen was diluted in a 1:5 relation with EYT extender (based on egg yolk and buffer Tris: 510 mOsm/L and 7,5 pH) or Test extender (based on egg yolk and buffer Tris and Tes: 555 mOsm/L and 7,7 pH). The progressive motility (PM) and live sperms (LS) were evaluated. Each ejaculate, once diluted, was maintained for 30 minutes at 25° C and then cooled at a rate of 0.33° C per minute until it reached 5° C. Then each was stored in a refrigerator at 4° C. The evaluation of PM and LS was done at 24, 48, 72, 96, 120 and 144 hours after refrigeration. The PM was higher (P<0.05) in the EYT extender at 24 hours, reaching values over 50 % at 72 hours from its refrigeration. The LS did not show significant differences when comparing the extenders at different times, however, a linear descent was observed through time was observed. Considering that both extenders contain egg yolk and the main differences in composition between them was in the kind of buffer, the observed differences in the PM, could be attributed mainly to the physical properties of the extenders under study. In conclusion, it is possible to preserve cat spermatozoa, refrigerated at 4° C for periods of up to 72 hours with an extender based on egg yolk and Tris. |
