Diagnóstico molecular de la Parvovirosis canina en perros geriátricos

El objetivo de este estudio fue determinar la presencia de parvovirus
canino en perros geriátricos que asistieron a consulta con
sintomatología gastrointestinal compatible con parvovirosis canina
en el periodo octubre a diciembre del año 2021 en una clínica veterinaria
del sur de Quito, Ecuador, mediante diagnóstico molecular (PCR), se
tomó muestras de heces del recto con un hisopo y se homogenizó (10 %
p/v) en solución ARN Latter. Las heces suspendidas fueron separadas
por centrifugación y se utilizó alícuotas de 200 μL de los sobrenadantes
para extraer el ADN. Para el diseño de cebadores y sonda del GenBank
fueron recuperadas secuencias de nucleótidos de la partícula viral VP2
de algunas cepas del CPV-2, las cuales fueron alineadas mediante el uso
del software Primer 3. Los números y las cepas usadas para la alineación
fueron: CPV2: CPV-b, M38245 y CPV-Northern, M19296; CPV2a: CPV-15,
M24003 y CPV-31, M24000; CPV-2b: CPV-39, M74849 y CPV-133, M74852;
CPV-2 Glu426 mutante: 56/00, AY380577. Para la identificación de la
región objetivo del ensayo se realizó por visualización de la alineación
de la secuencia para amplificar un fragmento conservado de 93 pb
dentro de las secuencias alineadas de VP2. Se realizó PCR en tiempo
real con un sistema de detección en tiempo real y estos datos fueron
analizados con el software detector de software MXpro Real time qPCR
(versión 3.0). La mezcla de PCR de 25 μL para la reacción contiene el
mastermix Q Script, los cebadores (laboratorio VetNAAT), Eva Green
y agua libre de nucleasas hasta completar la reacción. Se detectó el
virus en el 28 % de los pacientes geriátricos muestreados.

For the design of primers and probes from GenBank, VP2 nucleotide sequences were retrieved from some CPV-2 strains, which were aligned using Primer3 software. The numbers and strains used for the alignment were: CPV2: CPV-b, M38245 and CPV-Northern, M19296; CPV2a: CPV-15, M24003 and CPV-31, M24000; CPV-2b: CPV-39, M74849 and CPV-133, M74852; Mutant CPV-2 Glu426: 56/00, AY380577. Identification of the target region of the assay was first performed by visual sequence alignment to amplify a conserved 93 bp fragment within the aligned VP2 sequences. Real-time PCR was performed with a real-time detection system and these data were analyzed with MXpro Real time qPCR software detector software (version 3.0). The 25 μL PCR mix for the reaction contains the Q Script master mix, the primers (VetNAAT lab), Eva green and nuclease-free water until the reaction is complete. The virus was detected in 28 % of geriatric patients sampled. |
