Evaluación microscópica en conejos (oryctolagus cuniculus) de biocompatibilidad de aloimplante de meniscos conservados en glicerina al 98%

A fin de evaluar microscopicamente la biocompatibilidad de
aloimplante de meniscos conservados en glicerina al 98%, se
utilizaron 10 conejos machos de raza Nueva Zelanda Albina
(cinco como donadores y cinco como receptores). De los donadores,
cinco meniscos mediales frescos se utilizaron para estudio
histológico (grupo control) y cinco se conservaron en glicerina
al 98% por ocho meses para posterior implantación (grupo
experimental). Previo a la implantación, los meniscos conservados
se introdujeron 24 horas en recipiente plástico estéril que
contenía 11 mL de solución constituida por 10 mL de cloruro de
sodio al 0,9% y 1 mL de gentamicina (40mg) quedando a una
concentración de 4mg de gentamicina. Después de 60 días de
aloimplantación en rodilla de conejo, estos fueron sacrificados y
los meniscos se retiraron y prepararon para estudio histológico.
El estudio estableció que hubo crecimiento de células en los
meniscos post-implantados y que las medias del conteo celular
de los meniscos post-implantados presentaron diferencias estadísticamente
significativas con los pre-implantados más NO
hubo diferencias estadísticamente significativas entre las porciones
de un mismo menisco. Además, el tipo de fibra predominante
en los meniscos implantados fue colágeno tipo I en un
82% y el porcentaje restante (18%) correspondió a estructuras
celulares oxidadas con citoplasma, indicativo de degeneración
celular. Los meniscos aloimplantados en rodilla de conejo fueron
biocompatibles porque permitieron crecimiento de células
constitutivas de menisco (condroblastos, condrocitos, fibrocondrocitos),
pudiendo considerarse una opción viable de reemplazo
tisular en caso de lesión.

Resumen en otro Idioma | In order to evaluate microscopically the biocompatibility of meniscal alloimplantation in rabbits preserved in 98% glycerin, ten Albino New Zealand male rabbits were used (five donors and five receptors). Five fresh medial meniscus were used, taken from donors for histological study (control group), and five were preserved in 98% glycerin during eight months for late implantation (experimental group). Prior to implantation, the preserved meniscus were submerged during 24 hours in a sterile plastic container containing a 11 mL solution of 10 mL sodium clorure to 0.9% and 1 mL of gentamicine (40mg), leaving a 4 mg concentration of gentamicine. After 60 days of alloimplantation in rabbit knee, these were euthanatized and the meniscus were removed and prepared for histological study. The study could establish that there was a growth of cells in the post-implanted meniscus and that the averages of cell count of the post-implanted meniscus presented statistical meaningful differences compared with the pre-implanted. However, there were not any statistical meaningful differences among portions from the same meniscus. Also, the type of predominant fiber in the implanted meniscus was 82% of type 1 collagen and the other percentage (18%) corresponded to oxide cell structures with cytoplasm, an indicator of cellular degeneration. It was concluded that the alloimplanted meniscus in knee were biocompatible because they let the growth of constituting meniscus cells (chondroblasts, chondrocytes, fiberchondrocytes), being this considered a viable way to tissue replacement in case of injure. |
